Vertebrate Freeze Tolerance: Molecular Studies of Signal Transduction and Gene Expression

چکیده

Various amphibians and reptiles have the extraordinary ability to endure the freezing of body fluids while overwintering. In studies with the wood frog, Rana sylvatica, we continue to explore new facets of vertebrate freeze tolerance. New studies have analyzed modifications of wood frog insulin and the controls on protein kinase A and protein phosphatase-1 that regulate glycogen metabolism in order to determine how normal homeostatic control over glucose is overridden to allow glucose to reach the extreme concentrations needed for cryoprotection. The roles of mitogen-activated protein kinases in mediating cell responses to freeze/thaw are also being assessed. Other studies are using cDNA library screening to analyze freeze-induced gene expression. Novel results include the identification of genes, protein products and cell functions that have never before been implicated in natural freezing survival.

Strategies for exploration of freeze responsive gene expression: advances in vertebrate freeze tolerance.

Winter survival for many cold-blooded species involves freeze tolerance, the capacity to endure the freezing of a high percentage of total body water as extracellular ice. The wood frog (Rana sylvatica) is the primary model animal used for studies of vertebrate freeze tolerance and current studies in my lab are focused on the freeze-induced changes in gene expression that support freezing survi...

انتقال ژن‌های OF2 و VAP به چغندر قند با کمک آگروباکتریوم ریزوژنز برای بررسی مقاومت به نماتد

OF2 and VAP genes, probably involved in signal transduction of sugarbeet nematode resistance, have already been cloned in bacterial vector by AFLP molecular marker and a two-hybrid system, respectively. To examine their capability to introduce resistance in sugarbeet, the genes were transferred to plant expression vectors. For this reason, OF2 gene after isolation was inserted within T-DNA of p...
